You will need the latest iClone, I can promise you that (just the way it works - they release a version which the plugin needs.  I'd bet a dollar this won't work with anything less).

If that's a Python plugin I'd have to say it may well be as much as anyone needs unless they are some sort of architectural guru.

In my (admittedly limited) playing around with it, this BuildingGen could definitely be emulated with some Python code.  PLUS it could be simplified - I think the idea behind it is great, but it also feels like a beta thing that really needed more, well, beta testing.  But now that I've got my new machine setup I'm going to give it a huge try today and will post my results.

Is it possible to layout actual building plans /blueprints and build a 3D model in iClone with BuildingGen?

You do know you can download a trial version and play with it, right?

It's more of a modeling tool that can use modular resources to put together structures for use in animation.  No serious architect or other uses are intended (which isn't to say you can't get close to a lot of laid out plans, but I don't think your usage case is possible).

But try it yourself (or at the very least watch the video tutorials - won't take you more than 30 minutes to completely understand what it's all about).
