I can't help you with the rigging question--no idea if that's possible or how it would be done. But morphs can create changes in the mesh, like you're talking about with the wrinkles. I've done this to create some subtle changes in one of my characters' expressions and make him able to breathe and swallow. Simple version is I used the Mesh Editor or the head morphs to change the mesh in CC3, exported the various morph states as iAvatars, then brought them into iClone with the Morph Creator and animated them with the Morph Animator.

It'll try to make a video showing how I did it. The first time was hard, but now that I've had practice, it's super simple. 

Maybe some one else can answer the rigging question, but I'll do the part I can.

You can create custom blendshapes and animate them. It's on the modify pannel of the FaceKey.

But you need 3DXChange to access them on the faceKey panel. You add them in the 3DXChange Expressions panel.

For the second question , you can have access on the face bone by exporting your avatar in fbx format and play with on other 3D software but In the Iclone univers you can't have access.

If you're using UE4 in your workflow, this demo is basically what you are need - 

I'm sure something similar can be done with Unity, but I don't use it in my work so can't help you with this.
I will release the project files that I used to achieve this result for free in the next few days along with a video tutorial. Maybe I'll also create a forum post for more people to see, but I'm not sure yet which topic will fit this type of content.
